To start Rune Mysteries speak to Duke Horacio, you can find him in a room on the 2nd floor of Lumbridge Castle. Ask him if he has any quests for you, he will ask you to take a strange talisman to the head wizard of the Wizard Tower. Agree to help and he will hand you an air talisman.
Delivery-
Now head to the Wizard Tower, it is found south of Draynor, across a large bridge. Climb down the ladder into the basement and talk to Sedridor, who is in a nearby room. Tell him you are looking for the head wizard, then after a bit of talking agree to hand him the talisman. Sedridor will be amazed at the talisman and ask you to take a package to Aubury, who owns the rune shop in Varrock. You can find the shop directly south of the Eastern Varrock bank.
The Secret Revealed-
Talk to Aubury and tell him that you have a package for him. Aubury will talk for a bit then stop. Talk to him again and he will give you some notes to take back to Sedridor. Do this and Sedridor will decide to reveal the secret he and Aubury mentioned earlier. He will tell you that long ago wizards knew how to create rune stones, but that the knowledge had been lost. He will then explain the details of this technique and how you helped find it again.
